No, you can only use one column at a time to filter with the "Lookup Multiple Rows in Other Table" enrichment. This feature allows you to search a specific column in another table, but you can't apply multiple columns as a filter in your lookup.
The lookup action requires you to specify:
** Table to Search
** Target Column
** Filter Operator
** Row Value
